The demand for whey and plant based protein powder supplements continues to increase rapidly as consumers search for a quick way to meet their dietary needs. Accurate food authentication methods surrounding the protein content of these supplements is slim, therefore development of new methods proves essential. A method involving reversed phase high performance liquid chromatography (RP-HPLC) was developed as a simple way to analyze the 9 essential amino acids in whey and plant based protein powders. All amino acid standards went through a pre-column derivatization reaction involving 9-fluorenylmethyl chloroformate (FMOC-Cl) in a 50:50 borate buffer and acetonitrile (ACN) solution at room temperature. Standards and protein samples were prepared and run on the column individually for accurate results. Different filtration and extraction techniques were experimented for best preparation for the reversed phase column. This method was applied to samples of various brands of whey and plant protein powder.
