Diabetic wounds tend to heal slowly and can lead to long standing chronic wounds and amputations. Two interleukins, IL-17 and IL-20, are implicated in the pathophysiology of this aberrant healing process. IL-17 is a proinflammatory cytokine, while IL-20 induces keratinocyte proliferation. Suppressing these interleukins with antibodies has been shown to improve wound healing. However, antibodies may not be the best choice for developing shelf-stable wound treatments. Aptamers, particularly DNA aptamers, could be a preferable option due to cost and improved stability. Currently, there are no reported DNA aptamers for IL-17 or IL-20 and only an RNA aptamer for IL-17. Using systematic evolution of ligands by exponential enrichment (SELEX), we are isolating DNA aptamers for these interleukins. The outcomes of these selections and the characterization of the candidate aptamers will be discussed.
