Thiocyanate concentration in saliva has been suggested as a chemical marker for screening analysis for oral and gastric cancers. To capitalize on this, we have developed a chemosensor electrode used for the electrochemical detection of thiocyanate in an aqueous medium. The electrode is comprised of a fluorine-doped tin oxide (FTO) coated glass substrate that has been subsequentially functionalized with: a doctor bladed and cured anatase TiO<sub>2</sub> layer; silanation with ethnyltrimethylsilane; covalently bound iron (II/III) tetratolylporphyrin via Sonogashira coupling. The focus of this experiment is a characterization of synthetic saliva samples for proof of concept detection of thiocyanate in direct saliva and in dilute saline solution to mimic an oral rinse. Thiocyanate concentration standards will be prepared with sodium thiocyanate to form solutions between 0.1 µg/ml to 10 µg/ml along with unadulterated synthetic saliva serving as a control solution. Samples will be characterized by cyclic voltammetry both directly in the synthetic saliva solution and in solutions of synthetic saliva diluted with 0.1 M KCl (aq).
