Cisplatin and other platinum compounds are an essential part of combination chemotherapy to treat cancer. The compounds bind to and crosslink DNA, thus leading to cell death. Due to the high growth rates of tumor cells this treatment impacts cancer cells the most. While many patients exhibit high initial responsiveness to treatment, eventually many also relapse due to drug-resistance of the cancers. The reasons for this resistance are not well understood; the eventual failure of the drug poses a major hurdle to successful cancer treatment. Also, while it is known that crosslinking DNA-strands can lead to apoptosis, only few DNA-platinum adducts have been isolated and fully characterized. Many platinum-based drugs cause considerable side effects.<br/><br/>In this project, hexose platinum conjugates that are potential substrates of hexose transporters will be synthesized. The conjugates will be subdivided into three domains, specifically a hexose derivative as transporter substrate, a spacer, and a chelant for platinum.
