In early 2020 Pfizer initiated discovery efforts to identify an oral SARS-CoV-2 M<sup>pro</sup> inhibitor for the treatment of COVID-19, which resulted in the discovery and development of Paxlovid (nirmatrelvir [PF-07321332] tablets and ritonavir tablets). This was achieved against highly aggressive timelines aka ‘Lightspeed project’ and required all hands-on deck with lots of unconventional thinking. The R1 sourcing strategy for PF-07321332 from alpha raws to R1 Raws, GMP API, and non-GMP API for Reg Tox was critical in meeting these unprecedented timelines and mitigating risk associated with speed, highlights along with learnings from this work will be presented.
