Axially chiral compounds are common structural motifs in natural products and privileged ligands and catalysts. Among them, axially chiral indole derivatives received considerable attention due to their unique structures. However, asymmetric construction of such five-membered-ring atropisomers is quite challenging due to their relatively low rotation barriers. Meanwhile, transition-metal-catalyzed C-H functionalization has achieved great success in the asymmetric synthesis of axially chiral compounds, which is now regarded as a reliable method. Considering the importance of these indole-based atropisomers, several transition-metal-catalysts including Rh/Ir complexes were employed to promote C-H activation and control the enantioselectivity. In this talk, our recent works on asymmetric synthesis of indole-based atropisomers through transition-metal-catalyzed C-H functionalization reactions will be discussed.
