Aldosterone, the steroid hormone produced by aldosterone synthase (CYP11B2), is responsible for the homeostatic regulation of sodium and potassium levels in blood, and when upregulated can have deleterious effects on the body and cause hypertension, heart failure, and myocardial fibrosis. Previous studies have shown that inhibitors of aldosterone synthase effectively attenuate its production and are the preferred treatment of hyperaldosteronism over mineralocorticoid receptor antagonists (MRAs), which have been shown to cause adverse side effects. A greener method was employed to synthesize potential inhibitors using solventless aldol condensation methodology. One novel enone, (E)-2-[(4-pyridyl)methylidene]-5-fluoro-1-indanone, was synthesized and characterized via IR, NMR, and mass spectrometry. Computational comparisons with known CYP11B2 inhibitors were performed. Future work could involve evaluating the inhibitory activity and selectivity of the inhibitor towards CYP11B2.
