Antibiotic resistance is a growing health concern and has been deemed a global threat. Therefore, it is essential to develop novel drugs to combat the rise of multidrug-resistant bacteria. This project sought to focus on these issues by developing an adjuvant that has the ability to restore the effectiveness of already-existing antibiotics by increasing accumulation in Gram-negative bacteria, such as <i>Pseudomonas aeruginosa</i>. It has been established that poly-nitrogenous adjuvants can effectively cross the outer membrane of Gram-negative bacteria. Specifically, the presence of primary amines and guanidines increase the accumulation of antibiotics within Gram-negative bacteria and promote diffusion across the OM. Current efforts are focusing on the development and synthesis of cleavable antibiotic-adjuvant hybrids through covalently attachment of the antibiotic and adjuvant. From previous work, the bis-amine containing adjuvant is being utilized to evaluate increased efficacy against Gram-negative bacteria.
