Tissue engineering has been gaining tremendous momentum for the development of scaffolds for growth and replacement of diseased or damaged tissue. In this work, we designed novel bio-printed scaffolds formed from newly developed collagen mimetic peptides. First, computational studies were conducted to determine stability of the self-assembled scaffold. The scaffolds were designed by combining plant based glycosylated compounds that were conjugated with cardioprotective peptides, and can potentially mimic the constituents of extracellular matrix of cells. The scaffolds were bioprinted into cardiac patches of various dimensions. Mesenchymal stem cells were then seeded onto the scaffolds in the presence of growth factors.  We examined the growth, differentiation of proliferation of mesenchymal stem cells into cardiomyocytes. Flow cytometry was utilized to explore if specific cardiomyocyte markers were seen over a period of time, including alpha-myosin heavy chain and cardiac troponin T. The mechanical properties of the bioprinted cell-scaffold matrices were then examined using rheometry, while the morphologies were examined by SEM. Immunofluorescence assays such as troponin T assay and actinin assay were conducted over a period of 21 days to determine if sarcomere assembly occurs. Our results indicated that collagen mimetic peptides have the potential to be developed into functional bio scaffolds for biological applications.
