Leishmaniasis is a poverty-associated disease that causes enormous burden on the health care system of many underdeveloped countries, especially in East Africa, South Asia, and Latin America. The etiological agent of Leishmaniasis is the protozoan Leishmania spp. (Trypnasomatidae), and it is transmitted by Phlebotomine hematophagous sandflies. There are more than 2 million cases per year of leishmaniasis infections distributed among the three disease forms: visceral, mucous, and cutaneous; the former one being a life-threatening condition caused by the variant Leishmania donovani. The cell-surface glycocalyx, plays a pivotal role in the parasite’s survival and infectivity. Among the glycocalyx’s components, the lipophosphoglycan (LPG) is a major glycan which structure varies according to the species and its life cycle. It is composed of a glycosylphosphatidylinositol anchor, a core glycan, a phosphor-polysaccharide, and a capping mannose-rich oligosaccharide. Given its abundance and outer-cell exposure, LPG is an important pathogen recognition pattern. Hence, LPG is an attractive target for developing carbohydrate-based vaccine candidate against Leishmaniasis. In this report, we present our progress towards the synthesis of the capping tetra-saccharide to prepare conjugated vaccine candidates and detection tools against Leishmania donovani
