In this work, we developed new tri-terpene peptide conjugates for targeting over-expressed receptors of tumor cells and their mutants. Furthermore, we created mutant peptides of known tumor targeting peptides and examined if those mutations resulted in enhanced binding interactions with the receptors. Initially docking studies were carried out using autodock vina, followed by molecular dynamics and MMGBSA studies. The conjugates that showed the highest binding interactions were then synthesized and self-assembled under aqueous conditions. Results showed the formation of either nanofibers or nanovesicles based on atomic force microscopy. We then examined their interactions with two different cancer cell lines. Actin cytoskeletal assays revealed that the nanofibrous assemblies caused cell blebbing, and reduction in proliferation. Thus, we have developed new peptide conjugates that may be develop therapeutics for targeting tumor cells.
