Our lab has created a library of <i>E. coli</i> cell lines expressing GFP-preptin fusion with alanine substitutions. Biological characterization has been hindered by the difficulty in isolating the pure analogs after cleavage from the GFP tag. We report a new purification scheme utilizing affinity and ion exchange chromatography.
