Archaeosomes formed by the self-assembly of sulfated lactosyl archaeol (SLA) glycolipids are vaccine adjuvants that can boost immunogenicity and vaccine efficacy in preclinical models. These SLA glycolipids are made up of a sugar moiety and an archaeol fragment. Herein, we report the synthesis of SLA glycolipids using archaeols of different chiral purities and investigated the effect of stereochemistry on their adjuvanticity. It was found that all SLA archaeosomes induced strong but similar humoral and cell-mediated antigen-specific immune responses following immunization of mice, irrespective of the chiral purities of the archaeols used. These responses were comparable or better than those obtained using mimetics of approved adjuvants. Next, we synthesized derivatives of SLA by replacing the archaeol moiety with various alcohols to have more insights on the structural-activity relationship.
