Cancer is one of the leading causes of mortality worldwide. Current cancer therapies suffer from many limitations such as lack of specificity, cytotoxicity, and drug resistance. Thus, advanced strategies for cancer therapy are urgently needed. Nanotechnology is a promising approach, as nanoparticles (NPs) can increase the solubility, bioavailability, and permeability as well as reduce the toxicity of many potent therapeutics. Liposomes are biocompatible and the first nano-delivery systems that have been successfully approved for clinical applications. Herein, we report a liposomal NP for the combination delivery of topoisomerase (TOP) I and II inhibitors, camptothecin (CPT) and doxorubicin (DOX) respectively. TOP II is an important target in cancer therapy. However, the clinical efficacy of drugs targeting TOP II is limited due to drug resistance and the associated side effects such as cardiotoxicity. To approach this problem, we study a combination delivery of TOP I and II inhibitors using liposomal-NPs which may achieve synergistic effects and overcome drug resistance and cytotoxicity. Liposomal NPs are composed of 1,2-Dioleoyl-sn-glycero-3-phosphocholine (DOPC), cholesterol, and CPT/DOX therapeutics in the core.
