Carbohydrates play an important role in various signaling processes and their synthetic counterparts have emerged as promising tools to interrogate human biology and address current health needs. In this presentation I will describe how experimental and computational insights into the mechanism of chemical glycosylation led to a general method for saccharide synthesis broadly defined as the glycosyl cross-coupling. These studies include C-C and C-heteroatom bond forming reactions which proceed with high chemo- and stereoselectivity under standardized set of conditions for a broad collection of substrates. We then applied this technology to identify and optimize new anti-infective candidates with improved activities and PK/PD properties but accessible with significantly reduced synthetic effort.
