dsDNA is most known for its ability to code for both genetic and structural information. Applying this encoded information beyond translation and transcription has yielded a wealth of interesting materials and is the foundation of DNA nanotechnology. Beyond its secondary structure, dsDNA exhibits exquisite variety in its geometric shape and topological form, but we have not yet tapped the full potential of this feature as a tool to fine-tune and control the properties of materials on a molecular scale. As these topological forms influence virtually every DNA process, vast literature precedent dedicated to understanding the role of DNA topology in life processes would serve as the ideal foundation for a unique material. In this work we report the use of DNA topological isoforms to control material properties. Using bioreactor derived plasmid DNA, our topological precursors are unimolecular defined polymers making the subsequent investigation of how topology plays into material properties easier. This work investigates the formation and role of topology in both bulk materials and bottle brush polymers derived from double stranded DNA.
