Fluorescent nanoparticles are of interest for a wide range of applications in biology and biomedical science. Particles with surface-bound fluorophores may detach and confound nanoparticle tracking studies, complicate further surface modification, and alter interactions at nano-bio interfaces. Therefore, internally fluorescent, polymer-encapsulated nanoparticles are highly desirable. We have synthesized and characterized fluorescent dyes and semiconductor quantum dots for polymer encapsulation with the goal of optimizing biological tracking. We are especially interested in obtaining highly luminescent, stable particles while avoiding spectral overlap with 4′,6-diamidino-2-phenylindole (DAPI) and green-fluorescent protein (GFP) stains to enable co-localization imaging. We present synthesis methods, full characterization, in vitro and in vivo tracking, and toxicology data on these materials.
