Degenerative diseases, which currently remain without a cure, are linked to misbehaving proteins and peptides which undergo self-assembly and aggregation leading to cytotoxic structures. The proteins of interest are tau and TDP-43 which are linked to neurodegeneration, and gelsolin and crystallin which are linked to eye, skin, and liver degeneration. Using biochemical and biophysical methods we have discovered that all proteins make amyloidogenic structures with polymorphic morphologies. In addition, the aggregation can be inhibited by using small organic molecules or large antibodies, both of which are viable therapeutic strategies.
