Thiocyanate concentration in saliva has been suggested as a chemical marker for a screening analysis for oral and gastric cancers. In this project, we seek to adapt an existing technology for the electrochemical detection of thiocyanate in seawater to application as a screening tool for use as part of a routine dental exam. The manufacturing process of the chemosensory electrode begins with preparing a fluorine-doped tin oxide (FTO) coated glass substrate for the doctor-blading and curing of the anatase TiO2 layer. This is followed by silanation with trimethylsilylacetylene then Sonogashira coupling to attach a metalloporphyrin moiety. Here we optimize the TiO2 blading process by machining the process to consistently reproduce homogeneous TiO2 layers across multiple substrates. Additionally, designs of the sample compartment incorporate the chemically functionalized electrode along with a platinum wire counter electrode and a silver/silver chloride reference electrode have been optimized for different sample media – direct saliva samples or saliva diluted with saline solution consistent with an oral rinse collection strategy.
