Ongoing development of novel thio-based carbohydrate leaving groups creates methods for additional oligosaccharide assembly methods. The use of heterocyclic anomeric leaving groups containing nitrogens have been proven quite useful in the glycosylation reaction. This project outlines the synthesis and investigation of a nitrogen containing bicyclic carbohydrate leaving group, 4,2’pyridine-2-mercaptopyrimidine. This leaving group was affixed to common sugars producing novel glycosyl donors. These donors containing both ether and ester protecting groups were then subjected to glycosylations using a variety of thiophilic promoters. In addition to the approaches of promotion, glycosyl acceptors bearing a primary or secondary alcohol were also utilized.
